PGC-α and glucose metabolism in liver

My current research interest is focused on the anti-aging actions of the transcription co-activator protein, PGC-1α, a master regulator of energy metabolism. PGC-1α stimulates mitochondrial biogenesis, regulates glucose and fatty acid metabolism, regulates adaptive thermogenesis, modulates the fiber type composition of skeletal muscle and is essential for normal heart development. We have developed a transgenic mouse model in which PGC-1α is ubiquitously over-expressed. The phenotype of this model suggests that it has anti-aging actions similar to the anti-aging actions of caloric restriction and may therefore provide clues to the mechanism(s) of action of caloric restriction. In addition, the transgenic mice exhibit increased skeletal muscle insulin sensitivity. This observation is of great interest because one of the earliest symptoms of Type II diabetes mellitus, an age-related disease, is an increase in insulin resistance. The PGC-1α transgenic mouse model has the potential to provide insight into the development of insulin resistance and to identify target sites for pharmacological intervention.
